Biomimesys - Frequently Asked Questions

1. Basics of BIOMIMESYS

The BIOMIMESYS® hydroscaffold is the only technology for 3D cell culture based on Hyaluronic Acid (HA) crosslinked with other ECM (collagens, Fibronectin fragments, etc.). BIOMIMESYS® hydroscaffolds are provided in ready-to-use 96-well plates. With BIOMIMESYS® technology it is possible to better mimic the physiological cell microenvironment by modulating its composition, stiffness (Young modulus) and porosity. Thanks to HCS Pharma processes, the BIOMIMESYS®hydroscaffold is porous, and becomes translucent after seeding for microscopic observations.

The BIOMIMESYS® hydroscaffold is the only ready-to-use technology based on HA and different extracellular matrix components. With the BIOMIMESYS® hydroscaffold we can better mimic the microenvironment of each organ, healthy or pathological (composition and mechanical properties).

BIOMIMESYS® Adipose tissue, the collagen I and VI are mammalian origin and RGDS (Arg-Gly-Asp-Ser) is a synthetic fragment of fibronectin grafted on Hyaluronic Acid.

You can cultivate cell lines, primary cells and induced pluripotent stem cells (iPSCs) in BIOMIMESYS®. Different types of cells can be co-cultivated in BIOMIMESYS® (seeded at the same time, or sequentially; in the latter case a specific protocol has been developed. Please contact us for more information).

As soon as they are seeded, cells will enter the pores of the matrix. With time, your cells should colonize the hydroscaffold (under the form of spheroids for example). Since the hydroscaffold is composed of naturally occurring ECM components, cells will be able to remodel it as they would do in vivo.

Analysis techniques such as optical microscopy, immunocytochemistry, qPCR, Western blot, ELISA, histological techniques, electron microscopy, flow cytometry have been tested and are compatible with BIOMIMESYS®. All standard plates are in the 96-well format. If you do not need 96 hydroscaffolds, 96-well plates containing only 24 matrices are available. For specific formats (e.g. 6-, 12-, 24- and 48-well plates), please contact us.

For a first try, we recommend ordering the 96-well plate containing 24 hydroscaffolds.

BIOMIMESYS® hydroscaffold is porous, and the porosity depends on the model of the hydroscaffold: the values are indicated in each technical data sheet. We can also customize the porosity for your specific applications.

For commercialized models, the thickness of hydroscaffolds is below 800 µm. It can be customized for your preference.

2. How To Handle the Scaffold

It’s not possible to transfer the dry hydroscaffold. Be aware that if the hydroscaffold breaks, it will not be usable.

The wet hydroscaffold can be easily taken out from the wells, for example with pincers(forceps) or a pipette (with a gentle aspiration).

All hydroscaffolds from one plate should be used at the same time, otherwise the remaining ones will moisten and lose their properties. If you do not need 96 hydroscaffolds, you can order the plates containing only 24 matrices.

BIOMIMESYS® hydroscaffold plates are sterilized and put under vacuum: they are delivered dehydrated. It’s recommended to store the plates at 4°C. BIOMIMESYS® hydroscaffolds are stable for 12 months at 4°C. The date of release of production is indicated on the CoA of the product. We also guarantee reproducibility from batch to batch (another advantage of BIOMIMESYS technology).

You can hydrate hydroscaffolds and use them later by following the method described in the first step for co-culture protocol. However, be cautious with maintaining the sterility and other conditions of the hydroscaffold.

3. Selecting Biomimesys Models for Cells

You can cultivate cell lines, primary cells and induced pluripotent stem cells (iPSCs) in BIOMIMESYS®. Different types of cells can be co-cultivated in BIOMIMESYS® (seeded at the same time, or sequentially; in the latter case a specific protocol has been developed: please contact us for more information).

BIOMIMESYS Oncology is suitable for HT-29 cells.

For Keratinocytes cells we recommend to use BIOMIMESYS® Adipose tissue.

We advise to use BIOMIMESYS Oncology with colon carcinoma cells.

BIOMIMESYS plates are suitable for iPSC-differentiation. However, if the protocol was designed from 2D experiments, modifications of the concentration of growth factors might be necessary. The differentiation of iPSCs into hepatocytes is better in BIOMIMESYS Liver than in 2D, iPSC cultured in BIOMIMESYS liver showed much more mature phenotype in our laboratory.

For HUVEC cells culture we advise to use BIOMIMESYS® Oncology.

In BIOMIMESYS liver hydroscaffold, we advise to use plateable cryopreserved hepatocyte.

We suggest trying BIOMIMSYS® Oncology (collagen I) and BIOMIMSYS® Liver (collagen I and IV) and compare which one works better for your research. A customized hydroscaffold with collagen IV only is also available. Please contact customer support.

We are currently developing a new BIOMIMESYS® Brain hydroscaffold, for neuronal cell cultures (exhibiting neurite outgrowth in 3D). Neurons can also grow in current BIOMIMESYS hydroscaffold to form spheroids.

o We recommend using BIOMIMESYS® Oncology (collagen I). Or you can try BIOMIMESYS® Adipose tissue (collagen I and VI).

o Thanks to the physical properties of BIOMIMESYS hydroscaffold (solid), the in vivo implantation is possible without dissolution of the hydroscaffold.

BIOMIMESYS Oncology is suitable for HUVEC and Lymphoids cells.

We suggest trying BIOMIMSYS® Oncology (collagen I) and BIOMIMSYS® Liver (collagen I and IV) and comparing which one works better for your research. The customized hydroscaffold with collagen IV only is also available. Please contact customer support.

4. Cell Handling

BIOMIMESYS® plates should be taken out from the fridge (4°C) and unpacked just before use. The cells should be seeded within a small volume of medium (20-30 µL per hydroscaffold). Carefully dispense on top in the center of each hydroscaffold, without the pipette tip touching the matrix directly. The drop containing cells will be absorbed, and the cells will enter the pores of the matrix: BIOMIMESYS®’s appearance will change from opaque white (dehydrated hydroscaffold) to translucent. After you have seeded all hydroscaffolds in the 96-well plate, add the culture medium on the side of each well, with a final volume of 200 µL of medium and incubate the cells as usual at 37°C, 5% CO2. Note: if the seeded volume is too high, many cells will stay and grow out of the matrix. The ideal cell concentration of the cell suspension for each hydroscaffold should be determined by testing each case. Don’t hesitate to contact us to get information about cells we have already tested in BIOMIMESYS® in the past. For more details, please refer to the first step protocol.

Yes. It is normal to see bubbles following the cell seeding process. They should disappear within a few days, after changing the medium.

BIOMIMESYS hydroscaffolds have already been used (and are compatible) with the following assays:- EdU for proliferation assessment, Live/dead assay, Hoechst/Propidium iodide staining, Calcein green staining, XTT,WST-1. Other assays might also work.

We recommended hyaluronidase from bovine testes. Avoid toxic ones, such as bee venom.

o Thanks to the physical properties of BIOMIMESYS hydroscaffold (solid), the in vivo implantation is possible without dissolution of the hydroscaffold.

Seeding density, length of culture, and cell type are the known factors that determine the spheroid development.

In 3D, there is less cell proliferation compared with traditional 2D cell culture. Proliferation diminishes over time and is correlated with a reduction in the rate of spheroid growth.
